I wonder if regular rooted android ad blockers work on whatsapp.

Yes, as long as their ad servers are included in the dns blacklists.

remember when mobile apps had ads, and you could pay a 1 time fee of $0.99 or $1.99 to opt out of ads?

 Alright good enough for me, though I've noticed there are a lot more adblocking detection enabled apps these days.
