Does 4 gpus and a m.2 work at the same time?

Correct me if I'm wrong but I've kind of assumed M.2 connects over DMi 3.0 so however many direct PCI-E lanes you utilize, they'll never hinder M.2.

Well first of all, don't use 4 GPUs because it's a horrible idea.

An M.2 drive will connect to the chipset via four (I think) PCIe lanes, and then the chipset is connected to the CPU via DMI. I think the confusion is coming from the fact that the chipset has it's own set of PCIe lanes (8 lanes of PCIe 2.0 for X99, and 20 lanes of PCIe 3.0 for Z170) in addition to those from the CPU.

 

However, X99 systems still have DMI 2.0. DMI 3.0 is a Skylake feature. And I don't think four GPUs can be done on Skylake—at least not in SLI.

For gaming, 4 GPUs is a really horrible idea, unless for some reason the most powerful cards aren't strong enough for you, and even then, you'd have better luck getting a Flux Capacitor to work properly and grabbing a GPU from years later. 

 

For compute tasks, on the other hand, go bonkers. 3d rendering and other such tasks scale far better, and do not require the GPUs to be in SLI/Crossfire. 

 

From what I've read in my manual for my Asus board (H97 Plus), use of the M. 2 slot shuts off certain SATA connectors, though no further I'll effects than that.
