Yes. AIO watercoolers often have thermal paste preapplied

Thermal interface material is used to bridge the air gap between two surfaces when the intention is to conduct heat. if you could perfectly polish/lap both surfaces there wouldn't be a need for the interface material. Do keep in mind many modern coolers come with a pre-applied film of TIM, in this case there is no need to add more to the cold-plate.

OT:

Yes. The thermal paste is required when using any cooling solution. Always.

 

While you theoretically can run a CPU without it, there are a shitload of boxes that need to be ticked in order to prevent the CPU from overheating.
