If you're running Win7 or 8 chances are you're already doing so automatically.

Case: Meatbag, humanoid - APU: Human Brain version 1.53 (stock clock) - Storage: 100TB SND (Squishy Neuron Drive) - PSU: a combined 500W of Mitochondrial cells - Optical Drives: 2 Oculi, with corrective lenses.

I usually double check every month or so and defrag if the fragmentation is above something like 2%. Mostly it's just to feel better. If you leave your computer on all the time it is probably  being ran once a week automatically. If you don't then you will need to run it manually once in a while.

My rig: 2600k(4.2 GHz) w/ Cooler Master hyper 212+, Gigabyte Z68-UD3H-B3, Powercolor 7870 xt(1100/1500) w/AIO mod,

8GB DDR3 1600, 120GB Kingston HyperX 3K SSD, 1TB Seagate, Antec earthwatts 430, NZXT H2

Verified max overclock, just for kicks: http://valid.canardpc.com/show_oc.php?id=2609399

On my desktop I never do as its SSD for my OS and the other 2 HHD don't really need too, I am also running windows 8 so....

On my aging netbook I try to do it monthly about the same time I run CCleaner to tidy the drive up.

If you download Defraggler, you can set it to defrag automatically at set times, or on boot. If you move around stuff a lot, do it about once a month or so.

I don't defrag unless fragmentation is over 9-10%. After that it get noticable.
