SATA Hot Plug

So, in the BIOS, my H110 MB has an option to toggle one or more SATA ports as SATA hot plug. Does that mean I can connect and disconnect a SATA drive when I need?

Yes.

Hot-swap/hot-plug allows you to switch non-essential drives without having to shut down the PC.

should also note that theoretically the hard drive needs to support it as well, and that this is technically not meant for a drive you'll be connecting and disconnecting trough seperate sata and power cables.

So they will need to stay powered on, but I could, for example, disconnect an inactive disk reader, and connect a hard drive?

It sounds too simple.

the feature is meant for those hotswap docks to put in 5.25" bays, or for eSATA ports for external hard drives.

Also good to let the drive in cheap docks without a power switch to spin down before moving them. Also they can get pretty fucking hot to touch.

EDIT: should be aiming this @BackfiringOnMe

Silent build - You know your pc is too loud when the deaf complain. Windows 98 gaming build, smells like beige
