Is it ok if I applied thermal paste onto the CPU, install the cooler, then take the cooler out to check if i made a mistake, and after that put the cooler back on without removing and reapplying a new layer of thermal paste? will it create bubble? Given that  I haven't turn on the PC yet and it was within the time frame of 15-20 minutes or so.I

its probably ok but personally if you have more paste I would just re-apply a fresh layer

I wouldn't do that.

Why would you want to take off the cooler immediately after putting it on anyways?

as said above, it could be fine, tough it is highly recommended to apply a new layer of the stuff. 

if it was within 10/15 minutes of appying, and you did everything carefull enough, airbubbles in the compound should be minimal.

tough as airbubbles in this case are basically unavoidable, i would apply a fresh layer.
