I'm guessing two, because you didn't specify a chipset. There are B450, B550, X470 and X570 boards called ROG Strix E Gaming.

But SATA has B and M keys and NVME has one M key as much I know so are you confirm that both slots support NVME

It is common. I have the same on my B450-F Gaming. M.2_1 support both SATA and NVMe while M.2_2 support NVMe only.

EDIT: Beware. Some SATA ports may get disabled if you use M.2 drive. I don't know if this counts for Z390. Consult your manual on this.

Product page says so: ASUS ROG STRIX Z390-E GAMING | Gaming Motherboard | ASUS USA

Go there and scroll down to motherboard layout you will see this:

2 x M.2 Socket 3

‧ 1 x Type 2242-2280, supports SATA & PCIe 3.0 x4 modes
‧ 1 x Type 2242-22110, supports PCIe 3.0 x4 mode
