2nd PCIE slot

my friend has a h270 gaming 3 , he wants to know if he can use his 1080ti on the 2nd PCIE slot as he wants to connect something and if he puts the gpu on the 1st slot , the 2nd one is blocked from usage physically. 
Is it possible to be made without performance loss / with really low performance loss?

Technically there would be a bit (because electricity has to travel slightly further), but no big impacts should happen.

 

So long as they're both x16 slots he should be able to do it without noticing any performance loss.
