Yes, it will format the USB so you will lose all the data on there. 

Please quote our replys so we get a notification and can reply easily. Never cheap out on a PSU, or I will come to watch the fireworks.

you can create a partition on it  and should be able to keep the file

G'day, I have a 64GB which I've formatted for a Win10 install.

 

It came out as 32GB initially after I formatted, but that annoyed me because I wanted other files too (in my case drivers for my new PC).

 

I took the files off (all of them) re-formatted the USB back to 64GB, then put the files back and walla - an ESB USB with 54.5GB of 58.1GB (real GB) available.

 

I don't know if it'll work, but since I'm doing it myself I suppose I can recommend it (?). For safety, maybe just leave the default ESB if you're not impatient like I am, an don't have another PC to help you for troubleshooting/remaking the USB later if needed. Your call though.
