Yes, if you have hot-swap enabled on a port you're plugging it into.

Any unknown button should be pressed even number of times.

If you have hot swapping enabled on the respective port, giving it power and a data connection while the system is live is fine. That's why some cases have hot-swap features.

If the motherboard supports hot-swapping, it's a setting found in the BIOS somewhere.

I wouldn't recommend unplugging a HDD from the port because you will be moving it around while the platter inside is still spinning and this might cause problems. It is possible if you have "hot-swap" enabled in your BIOS and if the port supports it. 
Moreover, if you pick up a drive that spins at 7,200 rpm, the force from the spinning might force you to drop it and thus damage the data inside. I would be very careful not to damage the data if you value it. :)
