Serious Virus/Malware removal?

So earlier this week, I accidentally downloaded a virus, and it's been a huge problem since, I can't seem to get rid of whatever it is. I downloaded a video, that turned out to be some kind of virus, it was 400MB, I thought it was a video, I didn't even check before I double clicked it. Now, I've rebooted into safe mode, and run Microsoft Security Essentials, and Malware Bytes both, but it's still not gone... I don't know what else to do besides reformatting ,which I really don't want to do right now. Help!

AVG?

Someone told Luke and Linus at CES 2017 to "Unban the legend known as Jerakl" and that's about all I've got going for me. (It didn't work)

AVG and MalwareBytes. If one can't find it, the other will most likely. Usually its MalwareBytes that doesn't catch something AVG does.. Rarely it's the other way around but I've had it happen.

Also, second question....

 

If I had 3 HDDs, and I had Boot HDD 1, and I downloaded the virus file on HDD 2, should I wipe both? Even if I deleted the virus from HDD2

Well first off, how do you know it's a virus? What is happening on the computer that lets you know that MBAM and MSE haven't fixed it? There may be other options before you nuke the OS from orbit.
