The HDD itself will be fine, but it might end up damaging the SATA connector, which would be nearly as bad as killing the drive. SATA isn't made to be plugged in thousands of times like external connectors are (e.g. USB, 3.5mm audio jacks). It will definitely last more than the 50 cycle minimum stated by the spec, but if you're planning to use a HDD as a big USB drive consider either not doing that or getting an external enclosure.
