So PCPartPicker is telling me that the M.2 slot #1 on all of the Z590 boards I’ve picked is non-functional.  Is that true?

The nr1 M.2 slot on most Z590 boards only work with a 11th gen cpu

Okay, so it’s saying the M.2 slot won’t work because 11th gen CPUs aren’t available for selection on PCPP yet.  It’s assuming I’m using a 10th gen CPU.  That makes sense, right?

That slot is pcie 4.0 and you need an 11th gen cpu to make use of it. Looking on Gigabyte's website it just says supported by 11th gen only for that slot.
