Yes, you will want to have thermal paste between your CPU and cooler.

It just helps with optimal contact between the two, so the CPU can be properly cooled. If you're just running the CPU for a minute or two, I would not consider it necessary, but for normal operation you will want thermal paste.

The IHS(That smooth aluminum surface of the CPU) and CPU cooler surfaces are NOT perfectly flat and there are small imperfections in the surface. Thermal paste is used to fill in those voids and provides proper transfer of heat generated by the CPU to the CPU cooler. No matter if you are overclocking or not, you need thermal paste.
