it should do it when you uninstall it via the software it usually provides

Right click on the program you want to uninstall and uninstall it.

Should I then use unins000.exe to uninstall the game? it is in the game folder.

This is a question that could have been easily been googled, that being said depending on the game and where it is from i.e. Steam, EGL, Ubisoft etc there is an option generally speaking if you right click on the game that you are wanting to remove it will give you an option to uninstall it there. Just the easiest way to do it apart from what @Visherasaid.
