From the looks of modern computers, it seems that eSATA has been abandoned by most companies, but I got another BTX Dell OptiPlex SFF, and I noticed on the motherboard the eSATA port (Not on the back, but on the board), and while I don't plan to use the eSATA port, is there a differnce between regular SATA II and eSATA? Is there a reason why someone might use the eSATA port instead of a regular SATA port?

You can use that port for sata, its the same thing. The only differnce was the name in the bios.

I figured since a standard SATA cable plugs in. I wasn't going to use it, but I was just curious why it was there.
