So i can just put the second ram stick and it will automatically put the first ram stick to 1333?

If the motherboard has 4 memory slots, then the RAM should be in alternating slots (they're typically matching colours).
If it only has 2 memory slots then just stick it straight in and make sure it clicks and locks in to place as you push it in and you're good to go.

CPU: Intel i7 6700k  | Motherboard: Gigabyte Z170x Gaming 5 | RAM: 2x16GB 3000MHz Corsair Vengeance LPX | GPU: Gigabyte Aorus GTX 1080ti | PSU: Corsair RM750x (2018) | Case: BeQuiet SilentBase 800 | Cooler: Arctic Freezer 34 eSports | SSD: Samsung 970 Evo 500GB + Samsung 840 500GB + Crucial MX500 2TB | Monitor: Acer Predator XB271HU + Samsung BX2450

It only has 2 slots and the first slot is used by the 4gb so i just put it in the 2nd slot?

Yep.

Shut system down. Take the side panel off. Turn off PSU at the back of the unit and unplug the cord. Press the power button on the case again (the system will turn on for a second and you will see the fans start to spin up and then stop as there will be a little bit of power still in the capacitors of the PSU - this will drain it). The system will now be off. Insert the memory module in to the available slot. It should click in and lock in to place as you push it in.
Put the case side panel back on, plug the power cable back in to the PSU, flip the switch back on at the back of the PSU on, and turn the computer on.
Done.
