Would Mantle and DX12 optimize 8 cores?

Most games only use 4 cores.

Would Mantle and DX12 take advantge of i7s and FX8300s since they spread out threads more evenly, or would it take more time to see games truly taking advantage of 8 cores?

I agree. As far as I understand this current games are coded with these limitations in mind so running them on dx12/mantle wouldn't change anything. I don't think we will see any backports so most likely only new games will benefit from this.

Yep.DX12 will just improve the latency between the CPU cores and will increase the need for single threaded performance in order to feed the gpu faster.

Nothing to do with 6 or 8 cores optimization
