Yes, use 2 sticks.  Not much difference but slightly less stress on the CPU memory controller. 

4x8 single rank and 2x16 dual rank could perform about the same, but vs single rank difference is small.

 

also, using 4 dimms is harder on the memory controller so perhaps you would have to go lower on speed on 4 dimms

 

HOWEVER

 

4 dimms looks better than 2 in every scenario

If you're talking about DDR5, 2x16GB is the only thing you should consider. 

 

If you're talking about DDR4 and earlier, it's a lot more complicated, since there are a lot of factors to consider, the memory speed you're trying to achieve, the type of kit you're trying to buy, the motherboard you're trying to use, the memory controller you're using, etc. It becomes a question of memory ranks. The more memory ranks you've got, the more rank interleaving can go on, though usually after 2 ranks the returns are small at best. The more memory ranks you've got, the slower you can clock memory since there's too much load on the memory controller. Ryzen CPUs, for instance, top out at around 4800MT/s with single rank, 4000MT/s for dual rank, and ~3200-3466MT/s for quad rank (dependent on the specific chip). For a given achievable memory speed, you want the most ranks you can, but usually you want the fastest you can get overall (Ryzen is weird though and going above 3800MT/s usually results in performance regressions). 16GB DIMMs can be dual rank, especially if you go for really tightly timed ones like 3200MT/s CL14 or 4400MT/s CL17 since they're guaranteed to be Samsung 8Gb B die, but unless you're going out of your way to find dual rank sticks you're gonna get single rank sticks, so therefore 4x8GB will be faster for that speed, though if you just buy 2 2x8GB kits, you can run into issues with memory compatibility since you can run into mixed memory. 

 

2x16GB is usually the same choice, though 4x8GB, if you know what you're doing, can be a bit faster in some circumstances.

I would get the 2x16gb kit, and if you want to upgrade to 64gb later on you can get a second one in a few years.
