The retainer is just that, a mechanical retainer.

It's there to lock cards in slots so they don't push out if they're too heavy and sagging from the cooler weight. Some cases don't use screws to lock cards in slots, only pressure or rubber/plastic things going in the holes.. so with such cases it's possible for video cards to slightly pop out of the slot

 

If you screw the video card after inserting in position and you don't have the pci-e power cables pulling down on the card (just wire manage them to go through a hole slightly above the video card on directly to the right, instead of pulling it downwards) you'll be fine.
