What is the difference between these two storage? 

 

Samsung 860 Evo mSata 500gb

Samsung 860 Evo m.2 500gb

I'm buying for my nephew's motherboard it's a gigabyte b450i aorus pro wifi 

I'm not sure it's compatability

Then M.2 NVME is the way to go. That board apparently has both a x2 and x4 m.2 slot.

His board is a itx it's not that big

That doesn't mean he should waste money on an NVMe drive. There's no benefit as a boot drive. The 860 Evo M.2 is perfect.
