Just finished build and tried to flash the bios- which I started but after selecting the file on the usb stick and it installing- it reboots to ‘reboot and select proper boot device or insert boot media’ error!  Can anyone help as I worried I’ve screwed up badly!!!

I expect all is well.

Since you've done a flash and you're going to have to reconfigure might as well do a manual clear cmos just because.

Then I'm going to assume that your running Windows and that you need to configure the drive to boot uefi and use windows boot manager.

Try looking for a CSM setting in BIOS and setting it to uefi only.

It sounds like you don't have an Operating System installed on the machine yet. Install an Operating System and that message should be replaced with a beautiful boot screen of your OS of choice.
