If I use a NVMe M.2 SSD will I lose SATA Ports?

My motherboard has 8 SATA Ports, If I use a M.2 SSD will some of the ports stop working?

depends on the motherboard, check your mobo's manual or spec sheet to know.

On most, yes but only two of them when run at x4, while when you run at x2 you do not loose anything.
