Can apple time machine backup my mac and my external drive at the same time?

As long as the external HDD is partitioned in the correct scheme (MacOS Extended Journaled) then yes you can by adding the folder within the time machine settings pane.

FOr example, I have a macbook pro, and a 1TB external drives with movies. 

 

Can I set up a time machine drive (4TB external one) and use it back up my macbook pro as well as my 1TB hard drive?

Yes, actually you should be able to right in the settings, the same way you would use "Time Machine" to backup the mac you can also do the same with a external drive.

In theory (I have not tested) as long as the external drive is a Mac OS External (Journal-ed) file system format it should be auto mounted at the time of the back up, and automatically be backed up. There are options to exclude specific folders etc if needed.
