Which monitor? The one in the monitor is most likelyy for the built in speaker

Not all monitor have audio jacks and those jacks on monitors could be one of two things:

1. input

2. output

 

Some monitors have speakers inside and the jack can be used as input for your PC, so it can play your PC's sound on the speakers inside.

Otherwise it's output and a pair of speakers can be hooked up to the monitor, so the audio that comes through the monitor, can be put forward to your speaker set or headset. More commonly used with consoles, because those don't have jack outputs (well, now some of them do..)

 

Anyways, there is no 'preferred' output. Just use the one on your PC, more cables between you and the audio source might causes interference.
