I would back everything important from the drive up to another drive and then do some tests to see if the drive is healthy or not.
I would get a diagnostic tool from the manufacturer's website and run the quick and the extended tests and see if the drive passes them. It's also a good idea to get the raw values of the S.M.A.R.T. status as they will give you a more detailed information about the drive's health.
Defragging should improve the drive's performance if the data on it is fragmented. 
What's the drive's brand and model?
