OK to run 3x120 exhaust and 2x140 intake?

Do that and add another 120mm in the bottom as intake.  Then you'll be slightly positive for sure.

 

Technically, the area of the fans makes your current setup slightly negative, but I think the resistance of the radiator impedes those 2 exhaust fans enough to reduce their flow to make the system more balanced.

 

But since the radiator fans may spin independently of the other non-rad fans... I'd add the 3rd intake to be safe.

 

Edit:  Not sure if you changed it or were mistaken, but the r6 comes with a rear 140mm fan not 120mm.  Order another 140mm from Fractal and and bottom intake it, yes.

My plan for mounting the AIO in my build (Define R6) is to put the rad (240mm) on the top of the case as exhaust, and use another 120mm as rear exhaust. The case will have 2x140mm fans as intake. I've heard that the optimal configuration is balanced but slightly positive airflow. Will the 2 140mm intake fans be enough, or do I need to add a third? Is there another, better fan configuration I should use (8086k watercooled, 1080)?

I'm going to be changing out the rear exhaust to a 120mm because the fans I'm buying come in a 3-pack.

Great, thanks. One thing, the Fractal fans are not PWM and the other fans will be, so they won't be at exactly the same speed. Is this a problem? I guess I can manually adjust the curves to try and make them match up.

No, won't be an issue.  You'll be fine with that setup however you work it.
