I'm going to assume you want to upgrade the HDD in your Mac with an SSD. To my knowledge no you cannot exchange the data with just the two drives. You'd require a medium to temporarily store one while the other overwrite the first. Also I don't know what file system MacOS uses so even if you get a 3rd drive I can't be of help in the software department.

 

Personally I'm more partial to storing all my data on a dedicated drive or server so at any time I can dump my boot drive and reinstall the OS without catastrophic loss.
