I am looking into returning a possibly faulty motherboard, (asus strix Z270E Gaming Motherboard) and was just wondering on what exactly I had to return.

1. Do I need to put it back into the anti static bag? (if so rip I threw that out)

2. It came with a CD controlling the LEDs on the mobo, along with stickers and a coaster. Does all that need to be sent away as well?

3. What should the quality of the packaging be at returnal? (This is my first time ever returning an item that was purchased online, just wondering if theirs a special etiquette)

Not really sure on the anti static bag. I think they will need to shield for your CPU socket. I would say just call ASUS and ask from them what they require. That would be way faster to get the answer.

Bought from amazon

Since you're returning it to Amazon and not Asus directly, you technically don't need to have an anti-static bag or cpu cover. Include whatever accessories you have, but if you can't find everything, that's typically fine (with Amazon). Most of the time, Amazon return employees have no idea what the heck a motherboard is, or the components/accessories that need to be included. They wouldn't give less of a damn if you had an anti-static bag in there or not.

hmm I would have assumed so however quite a few people had been nagging me that I needed to at least have a cpu cover for an RMA. Not 100% sure yet.

An RMA is different than a return. An RMA means that you are sending your motherboard back to the manufacturer, (Asus in this case). The manufacturer will then attempt to repair your board and send it back to you. If they aren't able to, they send you back a refurbished unit.

A return is when you simply return the product to the retailer you purchased it from. Retailers are companies like Amazon, Newegg, etc. There are also manufacturer web stores where you can purchase the items directly. If you purchase an item from the manufacturer (like the Asus web store), it is best to include everything that you received and send the item back in undamaged condition. A return through Amazon or other retailers, however, will not mind if you do not include minor accessories or packaging material. This includes the cpu cover and anti-static bag. This is in part a result of most Amazon employees not knowing whether motherboards come with parts such as a cpu cover.
