You mean the backplate or the security lock lever that must be pulled to-the-side-then-down? About the backplate, it's risky because the cooler vibrations might cause damage on the long run. About the lever.. it should really matter. If it wasn't working, you'd already noticed it on the first boot. It's not gonna move afterwards if you have a cooler installed

Most, if not all, cpu coolers wouldn't fit to install if you left the lever up to secure the cpu. 

 

ZIF sockets go back to way before heatskinks were even necessary on cpus and they were all that held it in. If you forgot it, which I doubt, at this point if your computer is severely bumped when running it could cause an issue, but even that's questionable with the amount of pressure applied to the chip from the AMD hold down brackets.
