Depends on manufacturer as to if it voids. But most big brands, e.g evga allow you to install a waterblock as they have realised not doing so will alienate a sizeable chunk of the market.

Usually manufacturers will put little stickers on the screws or somewhere else on the card that you have to remove in order to get the heatsink off. If their isn't one you should be fine, just be careful not to destroy the thermal pads on the VRAM modules.
