I have a FiiO E6, which I use for my iPhone. Can anyone tell me if it would have any effect at all if I'd use this amp with my computer paired with a beyerdynamic dt990 pro 250ohm?

Just try it. It won't destroy any other device, that's for sure. So just set it up, and see for yourself the effect.

Then it doesn't matter. It's a subjective, personal taste. If you can't hear any difference, then just leave it at that, there's no difference (unless perhaps being louder)

For example: If I can't tell the difference of how a $10 steak tastes compared to luxurious $150 steak at a fancy French restaurant, then the difference wouldn't matter to me, right?
