Get three liters to be safe.

My personal loop which effectively has a thick 480 radiator and 1 cpu block with a ~200ml res took a full liter and the res is 3/4 full. Bearing in mind I still had some coolant in the rads so it takes roughly 1.5 liters.

EDIT: Bear in mind 2 liters may be enough.

EC6 Coolant should be replaced every year, correct?

Every loop should be serviced every 6-12 months. This includes a coolant change. So yes. But not only EC6.

I just know that some coolants should be replaced more often than others so that was my concern. So if I grab a drain port like this one: http://www.frozencpu.com/products/14596/ex-tub-1035/Bitspower_G14_Mini_Valve_-_Matte_Black_BP-MVV-MBK.html?tl=g30c101s460#blank and a T-Adapter: http://www.frozencpu.com/products/10382/ex-tub-622/Bitspower_G14_Matte_Black_T_Adapter_BP-MBTMB.html?tl=g30c499s745#blank

I just need to make a break in the loop and connect them with the T-Adapter and then screw on a compression fitting to the last non used side of the adapter and then get another one and slap it onto the valve and connect it. So when I go to drain all I have to do is screw on a compression fitting with a tube and drain it to a container of some sort? Does this seem right?

I do have a 500ish ml res, two 360 rads and 1/2" tubing and I have used about 1.4ish L of Coolant. Now I'd suggest you buy 3L and be good with it. I had a leak once, and then I had to have my PC left off a few weeks until the new coolant arrived. Rather have some backup, and with 3L you'll be able to fill it up and refill if necessary.

Any help with setting up a drain port properly?

Yes. All you need to watch out for is the drain port must be at the lowest point in the loop. Only things above it will drain out.

With a 765ml Reservoir + 2x RX480(60mm thick) rads how many liters of coolant should I look to buy? I was looking all over and I could find out how many liters a 60mm thick 480 rad consumes.

 I found that a colgate 480 radiator coolant capacity is 500ml (533.6 x 124 x 60mm)

That means my prediction was perfect. It will be just about 2L :D
