Yes you can.

I use Easy2Boot, which does not really have the best interface but it works rather well

Do you know how much room each of them take?  If not, go to MS's site where you're getting the ISO's and it will give you recommended sizes for each.

I have checked those sites. They say to have at least 4 GB of space. But is it for only 32 or 64 bit version? Or the combined? Cause in Windows 10 media creation tool there is a option to download both 32 and 64 bit ISO together.

My iso files are about 12 GB in total. How much space do you think I need in my USB drive? Will 16 GB be enough?

Also, will I be able to use my USB drive as a regular USB drive after creating a boot drive?

You need the space your files need, so yes...

You can put other files on it, it doesn't mind. It's going to be an ExFAT partition.

You can also tell the installer to leave some free space at the end if you want to make a completely separate parition.
