Can a router cause restriction of proxy use? specifically against wifi connections?

I can't get my proxies to work and I use a 2-in-1 from comcast and wonder if there are some settings that need to be tweaked in order to allow them.

No, the router or the wifi does not create any restrictions on proxys. You can go to the settings on your chrome browser to set your required proxy.
