Take the screw out and leave the standoff there if that is where the end of the m.2 SSD is after it's installed.  Then put the screw back in to hold down the end of the SSD.

But I don’t have any screws that fit into that standoff I was wondering If I really need that specific standoff or can I just put a screw in and replace it because it basically never touched the motherboard with the screw in

That looks like a screw in there to me.  And the standoff is there to make the SSD sit parallel to the board so if you have something else that can accomplish that then use it.  If you have a longer screw then you could probably just use that without a standoff if you only screw it down enough to make the SSD sit parallel to the board as it would had it been sitting on a standoff.
