These drives likely run through the Z370 chipset rather than using the cpu PCIe lanes. The bandwidth between the cpu and chipset is roughly equivalent to 4 PCIe 3.0 lanes. So there is a potential bottleneck if the two drives are saturating their NVMe connections.

I would think this depends on the expansion card. If the card can fit in a 4x slot, no. If the card can fit in an 8x slot, no. If the card only fits a 16x slot, I don't know, but likely no ;).
