The Mobo 8 Pin is different from the 8 Pin GPU connector, the pin shapes are different and should prevent you from plugging in the wrong one but if you force it I'm sure you could use the wrong one, better look closely before powering it on.

First off look at the connector from the power supply and the connector on the motherboard, each pin has either a square connector or D shaped connector in different order, I'll find a pic.

The PCI-E 8pin should have the word "PCI-E" written on the connector and some PCI-E 8 pins are 6+2pin which is easy to differentiate between this and the cpu 8 pin. (Corsair TX 650V2).
