That should be enough.  Ideally you want more intakes than exhausts.  Helps keep dust from seeping in through the cracks.

I am going to get the Define XL R2 case and was thinking about either the GTX 780 ACX, Twin Frozr, or DCu2, but i don't think that case has the proper airflow for those cards to get cool air so probably blower style would be better. I am thinking about getting 1 Corsair SP140 Quiet Edition for the front and 2 AF140 QE for the top two vents, but adding those might defeat the purpose of the case. It does come with a bottom mounted fan which could be enough for the video cards to get air, but im not too sure. Any Suggestions.

Blower style won't be better.  Theoretically less hot air goes into the case, but coolers with blower style fans are typically very mediocre.

 

As long as you have front intakes and top/rear exhausts, you'll be fine.  You don't need to worry too much about airflow.

Would adding just the one intake fan and not adding two top fans be enough? So essentially 3 intakes and 1 exhaust. I imagine that being inefficient.

Not too concerned cause the top 2 exhaust ports are moduvents all the intake fans have filtered intakes. Thanks for your answer. Looks like I'm just going to add the one intake fan in the front and if the temps aren't where i like it then add an exhaust or two.

I know that the Twin Frozr 3 is a blower style cooler. Having a bottom mounted fan as intake to the gpu should be able to keep the gpu cool.
