Yes, you can use it as a boot drive.

The difference would depend on the ssd and not just the form factor.

so the m.2 needs to be rated at a speed comparable to the pci-e connecter it attaches to to really see any speed improvement?

It will run at the speed its rated at which is normally around 500 mb/s and the bandwidth of M.2(Max amount of information it can transfer) is 10gb/s
