Yes, the data on the drive is duplicated including the OS. The connector used doesn't matter.

Im changing my laptop's internal HDD to SSD. Does the samsung data migration software copies the OS as well? Also, how do i know if my HDD needs a 2.0 or 3.0 USB port casing or does it matter?

You can also use a SATA to USB cable to transfer data from and to an internal drive with no need to buy a whole enclosure (like this one: http://i.imgur.com/rEidVFW.jpg). I do believe the Samsung Data Migration software moves the OS as well. You can also do a fresh install on the SSD and then transfer your data from the HDD to the SSD.

Either is perfectly compatible, but if your laptop supports USB 3.0 I'd recommend getting a 3.0 enclosure so you can take advantage of the extra bandwidth. A USB 3.0 hard drive will run at 2.0 speeds if that's all your laptop has, however.
