So two questions... 1 Can you RAID two drives of different capacities? and 2 can you RAID two drives after one has been in use for a few months?

Yes and Yes.

Raid 0 is when the data is striped across both disks, so should give the best performance but with the greatest risk. Whereas Raid 1 is mirroring one disk on the other. Raid 0 will probably work, but you'd only use 80GB of the 250 GB disk. You'd end up with about 160GB total space. To get full use of the 250GB disk, you'll need another 250GB hard drive for 500GB, for instance. 

 

Though I wouldn't.

Why not, if your RAID 0ing' two brand new quality drives such as Crucials, Samsungs or Intels your chance for failure isnt that bad

Well, you have answered your own question, haven't you?

This is only my opinion, you can do it as I said, but I wouldn't recommend it.

1) Yes, you can, but It is not recommend. You would not be able to properly stripe your data to both drives, what does this mean? Because you are using 2 different drive sizes, one larger drive will work harder since there is more data stored on it. This off-set will increase failure rate on the larger drive. If any drive fails from raid 0, you will corrupt and loose your data.

 

The best visual example I can give you, is a tandem bike, 2 people peddling 1 bike.

2) Yes, you can, again not recommended. The unknown hours on the used drive makes it difficult to accurately determine the Mean Time Between Failure (MTBF). MTBF of 2 new identical drives are the same, and can generally estimate the life of the array based on the hard drive's specification sheet.
