That path would define processes to exclude, but you're talking about files?

HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Windows Defender\Exclusions\Paths
HKEY_LOCAL_MACHINE\SOFTWARE\Microsoft\Windows Defender\Exclusions\Extensions

 

If Defender excludes something by default and doesn't let you remove it, I would assume removing it from the registry isn't going to work. It'll simply restore its defaults after a restart.
