Is it best to put the nvme boot drive in the first nvme slot or does it not matter?

It depends on your motherboard.  Generally the top m.2 slot on the face of the board is the primary for an m.2.

I have b550 Asus strix, so does that make a difference or no. Like I know the first one has the primary pcie busses but how much am I sacrificing if I put my boot m.2 in the second slot?

Ok, for your board, these are the specs of the top and bottom m.2 slots..:

1 x M.2 Socket 1 with M key, type 2242/2260/2280/22110 storage devices support(SATA & PCIe 4.0 x4 mode)

1 x M.2 Socket 2 with M key, type 2242/2260/2280/22110 storage devices support (SATA & PCIE 3.0 x 4 mode)

 

Nothing is going to be slow with that.. If it's not a pci-e 4.0 drive, it will be fine in the bottom slot.

It is worth noting that some older boards, will NOT boot if its not in the primary slot. That shouldn't be a problem anymore though.
