Some further context would be helpful.

 

A proportionality constant defines the relation between values which are connected by a proportion, so c in the equation y = cx. With definite sets of values for x and y, you just take corresponding values x1 and y1 and define c = y1/x1. If you have two random samples which are proportional to each other, then you find the proportionality constant through methods like linear regression.
