Is one mounting screw enough for a GPU?

It's fine. DOn't give it a second thought.

I had to reseat the GPU and ended up stripping the lower mounting hole. The top one is secure, and the card has an anti-sag bracket, but the lower screw is not snug because the hole is stripped. The card seems stable enough though. 
 

Should I leave it? I could try a wider screw if need be.

the bracket should provide enough support, i would not travel with the installed gpu tho

Would I use the spring washer on the loose screw? It isn’t quite “hand tight,” but at least it doesn’t fall out. Cheers!

You probably need a small nut to screw into. It's not necessary because you have a GPU support bracket. That is supporting the weight of the GPU, so those side screws are mostly to keep the GPU from popping out of the PCIE socket and to electrically ground the GPU.
