I plan to use a single ram stick of 16gb and my motherboard has 2 ram slots. Will it matter if i put the single ram stick on either of the two?

Check the manual, they usually have something in it which says which you should use first.

Motherboards with 4 slots usually recommend using slot 2 first, then slot 4, then the rest. I don't know how 2 slot boards handle this

That's massively dependent on your workload. It's not a necessity to get two (matching) RAM modules.

It'll likely just boot right up and work. However the motherboard manufacturers in the handbook recommend using A1. This slot is closest to the Cpu and will have the shortest tracings to the processor and less likely to have issues and probably the reason they recommend that slot.

If you have only one stick, there is literally zero difference what slot it goes into. It will get read from and written to all the same even with the less than an inch distance difference in electricity crossing a PCB.
