If you have only one stick, there is literally zero difference what slot it goes into. It will get read from and written to all the same even with the less than an inch distance difference in electricity crossing a PCB.

Check the manual, they usually have something in it which says which you should use first.
Motherboards with 4 slots usually recommend using slot 2 first, then slot 4, then the rest. I don't know how 2 slot boards handle this

That's massively dependent on your workload. It's not a necessity to get two (matching) RAM modules.
