I’m running a GTX 1060 in the chipset PCIe x16 (PCIe2 x4 electrically) slot on a mATX b450 and I’m not seeing any loss in performance. Mind you this is under Linux so it might handle the bus arbitration better than Windows.

SLI is not required nor recommended, as the cards are often doing different projects, and to the best of my knowledge cannot be configured to work together to fold the same protein.
 
On that note, I wonder how an R9 295x2 would fold, since it has 2 GPU's on the same PCB. Would that show up as two different GPU's? Would they too have their own work units assigned to each core? Would the heat output accelerate global warming by 57%?
 

Don't think SLI make any difference for folding

I made a another modification and added another GTX 680. I just don't know if I should SLI them.

We are producing more folding atm per week than we usually do in a month

I have a ryzen 2600 and a gtx 1660ti on an msi x570 prestige. i'm waiting for the 3950x and ek to make a monoblock. I'm also considering an rx 5700xt. I have big plans for Xerxes 2.0 =^_^= ...it's in beta for now... and the giant fan is for the folding. fans and watercooling stuffs coming soon.

Been busy and have not been on. But as promised, here is what I am running:
1. Pentium G630 with Radeon HD 68501
2. I7-4790s no GPU because the HP small form factor will not allow it.
3. I7-4770k with MSI Sea Hawk X 2080ti and I just installed a GTX 680 a few minutes ago.
4. I7-9700k with MSI Sea Hawk X 2080ti (only 1 GPU slot on my MB).

I am actually running 4 old machines that are 4700K's with no video cards. I also have 1 machine that is a 9700K with a 1080ti. They are all running on Full, 24 hours a day. I have noticed my electric bill is a a bit higher than normal. LOL.
