Typically, you can right click on the USB drive, click format, and choose the type of file format that you would like. If you're using it across OS's, then use exFAT (preferably, massive max file size) or if that's not possible, FAT32 (maximum file size of 4gb). If you're only using it on Windows machines, NTFS is a journaled file system that won't be as easily corrupted as the previous two.
Leave the capacity at max, and don't touch the allocation unit size, it's fine.
